چکیده

Asthma is a chronic inflammatory lung disease that causes respiratory difficulties. Black ginseng extract (BGE) has preventative effects on diseases such as asthma. However, the pharmacological mechanisms behind anti-asthmatic activity of BGE remain unknown. To investigate mechanism BGE, phorbol 12-myristate 13-acetate plus ionomycin (PMA/Iono)-stimulated mouse EL4 cells and ovalbumin (OVA)-induced mice with allergic airway inflammation were used. Immune (eosinophils/macrophages), interleukin (IL)-4, -5, -13, serum immunoglobulin E (IgE) levels measured using an enzyme-linked immunosorbent assay. Inflammatory cell recruitment mucus secretion in tissue estimated. Protein expression was analyzed via Western blotting, including inducible nitric oxide synthase (iNOS) activation protein kinase C theta (PKCθ) its downstream signaling molecules. decreased T helper (Th)2 cytokines, IgE, secretion, iNOS inflammation, thereby providing protective effect. Moreover, major ginsenosides inhibited production Th2 cytokines PMA/Iono-stimulated cells. In cells, these outcomes accompanied by inactivation PKCθ transcription factors, nuclear factor activated (NFAT), kappa B (NF-κB), activator 6 (STAT6), GATA binding 3 (GATA3), which are involved inflammation. also abovementioned transcriptional factors These results highlight potential useful therapeutic agent for

Human sulfatase 1 exerts anti-tumor activity by inhibiting the AKT/ CDK4 signaling pathway in melanoma

Human sulfatase 1 (hSulf-1) has aryl sulfatase activity. It can reduce the sulfation of cell surface heparan sulfate proteoglycan (HSPG) and inhibit various growth factor receptor-mediated signaling pathways. In most cancers, hSulf-1 is inactivated, which endows cancer cells with increasesed cell proliferation and metastatic activities, inhibition of apoptosis, and decreased sensitivity to radi...

Inactivation of mitogen-activated protein kinase signaling pathway reduces caspase-14 expression in impaired keratinocytes

Objective(s):Several investigations have revealed that caspase-14 is responsible for the epidermal differentiation and cornification, as well as the regulation of moisturizing effect. However, the precise regulation mechanism is still not clear. This study was aimed to investigate the expression of caspase-14 in filaggrin-deficient normal human epidermal keratinocytes (NHEKs) and to explore the...

Reversing multidrug resistance in hepatocellular carcinoma cells by inhibiting extracellular signal-regulated kinase/mitogen-activated protein kinase signaling pathway activity

The aim of the present study was to evaluate whether downregulation of extracellular signal-regulated kinase 1/2 (ERK1/2) is involved in conventional reversal methods and whether the inhibitors of the ERK signaling pathway reverse multidrug resistance (MDR) in hepatocellular carcinoma (HCC) cells. The sensitivities of SMMC7721 and BEL7402, and the MDR SMMC7721/Adriamycin (ADM) and BEL7402/ADM H...
